Taq Ligase is a thermostable DNA ligase that was cloned into E. coli cells from the thermophilic bacteria Thermus aquaticus.

Taq DNA Ligase stable and active at high temperatures

This enzyme can be stable and active at considerably higher temperatures than traditional DNA ligases, and it may be utilized in PCR conditions. The enzyme’s function is comparable to that of Tth ligase from related thermophilic species Thermus thermophilus.

It catalyzes the formation of a phosphodiester bond at a nick junction on double-stranded DNA. This enzyme does not tolerate any mismatches around the nicked region. Even a single-nucleotide mismatch reduces the ligation activity drastically. The unique specificity requirement of thermostable ligases makes them unsubstitutable in many advanced molecular techniques.

NAD+ dependent ligase from Thermus aquaticus (Taq DNA ligase)

Taq Ligase is a 74 kDa Thermus aquaticus ligase protein catalyzing the NAD+ dependent ligation of adjacent 3-hydroxyl and 5-phosphate termini in duplex DNA structures.

It doesn’t display activity on blunt ends or RNA substrates. This enzyme works in a wide range of DNA polymerase buffers with pH levels of 7-8.

Thermostable Taq DNA Ligase Applications

Ligases from thermophilic bacteria show considerably higher fidelity of even one up to two orders of magnitude more than bacteriophage T4 DNA Ligase.

Taq DNA Ligase can be utilised for a variety of applications including high-fidelity gene synthesis from overlapping oligodeoxynucleotides, Gibson assembly, repeat expansion detection (RED), multiple site mutagenesis, ligation amplification (LCR) targeted inverted repeat amplification, next-generation sequencing (NGS).

However, due to insufficient activity at low temperatures where 2- and 4-base cohesive ends form stable duplexes or blunt ends, it cannot be utilized to substitute T4 DNA Ligase in most cloning methods.
