Next-gen automated tissue cytometers
TissueGnostics manufactures fully integrated cutting-edge tissue cytometers for the scanning and the follow-up quantitative analysis of IHC, IF processed slides, tissue microarrays (TMAs) as well as cultured cells in well plates or cell culture flasks, and has a worldwide network of distributors making its advanced technology easily accessible.
The fully automated tissue cytometers engineered by TissueGnostics are available in multiple configurations for brightfield, fluorescence, multispectral, confocal, as well as high-throughput whole slide imaging (WSI) and include solutions for biomedical image analysis and quantitative pathology.
Fully automated and modular tissue cytometry platform
The TissueFAXS line imaging systems by TissueGnostics are modular and upgradable. Each imaging system is designed either in an upright configuration for scanning slides only or inverted for scanning slides and cultured cells in well plates or cell culture flasks.
All TissueFAXS systems come with an 8-slide stage, or a high-throughput automatic 120 slide loading system, available for the upright TissueFAXS system configuration. The TissueFAXS imaging systems can be equipped with high powered LED light engines combined with multi bandpass filter cubes for high-speed fluorescence scanning.
The TissueFAXS platform contains solutions for quantitative image analysis (HistoQuest, TissueQuest and/or StrataQuest).
From single-cell analysis to contextual tissue cytometry
TissueGnostics’ image analysis software is available in three forms:
TissueQuest for fluorescence single image analysis
HistoQuest for brightfield single-cell analysis
StrataQuest for multicellular contextual tissue analysis for both brightfield and fluorescence images
TissueQuest and HistoQuest are streamlined for rapidly acquiring nuclear segmentation and marker quantification per cell as well as dot detection and basic area measurements.
StrataQuest is a fully capable image analysis development platform, which can be used either to create unique analysis pipelines from scratch or predefined and standalone analysis algorithms called StrataQuest Apps. StrataQuest utilizes classical image analysis methods and engines as well as machine learning models and deep learning neural networks. StrataQuest Apps are packaged into simplified user interfaces which are comprised of macros, thereby enabling researchers with little or no experience in image analysis to obtain comprehensive high dimensional data from analyzing their scanned images.
All TissueGnostics software can be used standalone and is fully compatible with images produced by most scanning systems.
Cellular and tissue-based cytometry applications
The versatility of the tissue cytometry systems can support multiple cellular and tissue-based applications throughout the biomedical sciences, research and pharmaceutical development fields, including:
- Immunophenotyping – determine the immune status of organs/tissues in-situ (tumor biology, autoimmune disorders, allergies, transplantation immunology, etc.)
- Tumor & Cellular Microenvironment – perform phenotypic and functional characterization of cellular sub-populations in a spatial context
- Identification of single cells – identify single cells even in dense tissue, such as lymphatic organs through Deep Learning algorithms
- Automated classification of tissue structures – classify tissue structures trough Machine Learning into any number of classes per sample
- Multiplexing and spectral unmixing -perform high-plex assays on histological sections with single-cell precision
- Structural analysis and subcellular localisation – structural analysis and determination of subcellular marker localization
- Single-cell molecular profiling – Molecular single-cell profiling using antibody labeling, FISH, CISH, RNA-ISH, RNAScope, expansion microscopy – virtually any staining technique that can be visualized by optical microscopy
- Quantification of cellular pathogens – including intracellular parasites (e.g. leishmania) and viral load (e.g. SARS-CoV-2, Influenza, HIV, Zika, Dengue, Hepatitis, HSV, Chikungunya)
- Panoramic imaging and quantitative histopathology – experience fully integrated panoramic imaging of slides (even oversized slides) and whole slide quantitative analysis
Single-cell tissue imaging cytometry for preclinical studies
Preclinical in vitro tissue-based studies are critical in defining potential biomarkers and an integral part of lead candidate development. Flow cytometry is a well-established technique for preclinical research to profile therapeutics, including cell proliferation analysis and intracellular cytokine staining. Whilst it allows multiple biomarkers to be studied at one time, it has the downside of dissociating the cells from important information about the tissue microenvironment.
TissueGnostics aims to bring phenotypical and functional analysis as known from the flow cytometry extension fluorescence-activated single-cell sorting (FACS) into tissue context, but on a single-cell level.
This method has many advantages including, eliminating the need for a segmentation method at the cost of losing info on the original morphology of the tissue, being able to objectively quantify cell biomarkers, providing high statistical strength with very small-sized experiments highlighting imaging cytometry as a particularly cost-effective method, and overall significantly enhancing discovery of new therapeutics.
