Protean offers Thermostable Bst DNA Polymerase (Exonuclease Minus) ideally suited for isothermal DNA amplification, LAMP, WGA, MDA, RCA, CPA and other applications.

Bst DNA Polymerase is a recombinant 67 kDa Bacillus stearothermophilus DNA Polymerase protein with a 5′-3′ polymerase activity. It also has strand displacement activity and reverse transcription activity.

Thermostable Bacillus DNA Polymerase

The Thermostable Bst DNA Polymerase, exonuclease minus, shows a 5′-3′ polymerase activity, strand displacement activity, and reverse transcription activity. It has no detectable 3′-5′ exonuclease activity making its structure more compact and able to tolerate higher temperatures. The Bst DNA Polymerase concentration is 10 U/ul.

Main application areas include isothermal DNA amplification, Loop-Mediated Isothermal Amplification (LAMP), DNA strand displacement amplification, whole genome amplification (WGA), sequencing of DNA with high CG content and problematic secondary structures, rapid sequencing and amplification from very low amounts of DNA templates (10 fg).

Thermostable Bst DNA Polymerase ideal for LAMP, WGA and MDA

Bst DNA Polymerase, Large Fragment is a moderately thermostable enzyme from Bacillus stearothermophilus cloned to Escherichia coli bacteria. It displays thermophilic reverse transcriptase activity and is effective over a wide range of reaction buffer conditions and concentrations of magnesium ion (Mg2+). This polymerase shows powerful strand displacement abilities making it an ideal candidate for isothermal amplification (LAMP), whole genome amplification (WGA), and multiple displacement amplification (MDA).

With LAMP currently seeing increased use in molecular diagnostics as an alternative to PCR-based methods, because it does not require thermal cycling, the Bst DNA Polymerase (Exonuclease Minus) supplied by Protean is the ideal enzyme to answer the needs of researchers for amplification of DNA under isothermal conditions. A simple visual detection can be performed using fluorescence signal.
