Hyaluronic Acid Hydrogels

Thanks to their unique characteristics such as biocompatibility, biodegradability, flexibility and softness, in recent years, hydrogels built from Hyaluronic Acid (HA) have been studied intensively for biomedical applications such as cell therapy, tissue engineering, tissue regeneration, drug development, gene therapy and diagnostics.

Hyaluronic acid (HA) is one of the most commonly utilized biopolymers in the formulation of hydrogels designed for biomedical applications because of its biocompatibility, natural biofunctionality, biodegradability, non-immunogenicity and versatility. HA is a natural linear polysaccharide made of alternating units of d-glucuronic acid and N-acetyl-d-glucosamine, linked by β-1,3- and β-1,4-glycosidic bonds.

Hyaluronic Acid Function in the Extracellular Matrix

Because of its high molecular weight and its capacity to absorb a high quantity of water in the extracellular matrix of most tissues, HA helps to sustain the mechanical integrity of the tissue, homeostasis, viscoelasticity and lubrication.

Hyaluronic acid also plays a key part in intracellular functions. As it can bind to cell surface specific receptors (such as CD44 or RHAMM), HA regulates cell adhesion, migration, proliferation, and differentiation and, consequently, processes like inflammation, wound healing, tissue development, morphogenesis, tumor progression and metastasis.

3-D Life Hyaluronic Acid Hydrogels for 3D Cell Cultivation

3-D Life Hyaluronic Acid Hydrogels are biochemically defined hydrogels that may be used to cultivate a variety of cell types in three dimensions. The versatility of cell culture applications is enabled by the ease of use and full control of bio-molecular modifications and gel stiffness.

The hydrogel is made by combining thiol-functionalized hyaluronic acid with thiol-reactive dextran or polyvinyl alcohol (PVA). The presence of cell adhesion molecules (such as RGD Peptide, Cat. No. 09- P-001) in the hydrogel based on HA enables the spreading and migration of most of the cells.

For optimal cell compatibility, the hydrogel components are mixed at physiological pH (pH 7.2). The 3-D Life Dextran-HA Hydrogel and 3-D Life PVA-HA Hydrogel Kits also feature slow gelation kinetics that provide sufficient time to manipulate the solution before the start of gel formation.
