Recombinant Proteins

Recombinant HLA Molecules

Dyemer represents a breakthrough in T cell staining. Engineered from MHC class I molecules expressed in mammalian cell culture, Dyemers ensure proper glycosylation for biological relevance. These ready-to-use T Cell Staining reagents allow the quantification and enumeration of antigen-specific CD8+ T cells using flow cytometry. By multimerising peptide-loaded MHC class I complexes onto fluorophore-labeled streptavidin, Dyemers enable surface staining for precise detection.
myDyemer offers flexibility by allowing researchers to load specific peptides onto pre-multimerised MHC class I molecules. The myDyemer technique provides exceptional convenience and a fast experimental setup. Peptide Binding Analysis Services
The imuPep Class I Peptide Binding Analysis provides a cutting-edge solution to confirm the stability of peptide/HLA class I complexes. This service enables the assessment of immunogenic peptides swiftly and accurately, utilising a comprehensive array of recombinant HLA class I alleles.
Recent research has emphasised CD8+ T cells’ response to novel vaccines using peptides from in silico studies. However, the stability of the resulting peptide/HLA class I complex can remain uncertain. The complex’s stability can significantly impact peptide immunogenicity. Thus, a reliable assessment of complex stability is crucial during the early stages of developing new T cell epitopes.
imusyn has devised a novel method to load recombinant soluble HLA class I proteins with peptides. Peptide binding to HLA class I proteins is determined by Differential Scanning Fluorimetry (DSF), which measures fluorescence changes during temperature ramps. The peptide binders for a certain allotype can be ranked by their denaturation temperature (Ti), hence giving insights into the relative stability of the peptide/MHC complex. If DSF alone does not provide a conclusive result, peptide binding is confirmed by running the loaded complex on a native PAGE.
