Zero-step RT-PCR probe-based 2x Master Mix for RT-qPCR
The unique RT-PCR Master Mix does not require an isothermal reverse transcription step. Immediate cycling is made possible through a hotstart aptamer and a truly thermostable Taq DNA polymerase with reverse transcriptase activity, engineered by myPOLS Biotec.
Direct RT-PCRs are now also made possible thanks to the Volcano3G® RT-PCR Probe 2x Master Mix. As immediate hot PCR cycling parameters can break cell- and virus membranes there is no need for sample extraction and sample lysis.
Moreover, the Volcano3G provides superior detection of mRNA down to a few total copies. The increased brightness of the fluorescent signal provided by this Master Mix allows a convenient and accurate quantification.
Accelerating in vitro diagnostics
The Volcano3G® RT-PCR Probe 2x Master Mix is ready-to-use and contains an optimised reaction mix designed to achieve sensitive and reliable results. In many applications, sample extraction and sample lysis are not necessary. Additionally, as the hotstart aptamer formulation prevents unspecific amplification at lower temperatures (<57°C), a fast start function is implemented. This can significantly shorten laboratory time and accelerate the output of reliable in vitro diagnostics.
In addition, there is a pipetting aid in the form of a blue dye which will help ensure better visibility during aliquoting into well-plates. Because of Volcano’s thermostability, the entire PCR mix can be heated up at the beginning of the RT-PCR which is, together with the detergents in the mix, enabling immediate lysis to nonextracted virus, cell or bacterial samples.
The Volcano 3G Master Mix not only offers superior performance but also a variety of applications for precise and rapid qPCR.
Volcano3G shows an improved detection of mRNA down to a few total copies, which is clearly distinguishable from the No Template Control (NTC). The brighter fluorescent signal provided by the Volcano 3G Master Mix enables an easy and precise quantification.

Note: The Volcano3G® RT-PCR Probe 2x Master Mix is a unique master mix. If you are using it for the first time, myPOLS Biotec recommends designing primers to have higher melting/annealing temperatures (above 65°C). Additionally, for assay setup it is advised to perform RT-PCR test reactions with temperature gradients (i.e., for the annealing and extension steps) to identify the most proficient PCR protocol for your requirements using the companies Volcano DNA polymerase products.
