5D In-vitro Models Based on a Novel Fluidic Platform
IVTech is revolutionising in-vitro testing with its novel fluidic platform for groundbreaking 5D in-vitro models. These models go beyond 3D cultures by incorporating two additional dimensions. They combine 3D in-vitro structures, enriched by a dynamic environment, representing the ever-changing conditions within living organisms (IVth dimension), as well as an interconnected network to study cross modulation between tissues in a dynamic environment, where information transfer is facilitated by liquid exchange (Vth dimension).
Dynamic environments can mimic bodily movements like blood circulation, fostering more realistic cell culture conditions, whilst interconnected models provide valuable insights into tissue interactions. Utilising IVTech’s fluidic platform, researchers can create 5D models that are game-changing for permeability studies, bioavailability, toxicity testing, and multi-organ disease modeling.
Ready-to-use Kits for 5D In-vitro Models
IVTech offers ready-to-use kits comprising a peristaltic pump, an assortment of cell culture chambers, and advanced environmental control units. Researchers can apply familiar cell culture protocols standardised for multiwell or transwell plates like the classic 6, 12 and 24-well formats, as the cell seeding areas align with industry norms.
The transparent IVT chambers also allow cells to be monitored in real-time using an inverted microscope, enabling live imaging of cellular responses and tissue dynamics. The use of standard protocols and procedures coupled with the advantages of a cutting-edge fluidic platform represents a pivotal advantage for researchers.
Bioreactors for Physiologically Relevant 5D In-vitro Models
IVTech offers three main bioreactor suites, LiveBox1, LiveBox2 and MultiDyn.
LiveBox1 is a transparent bioreactor designed for dynamic in-vitro models in attached cell culture conditions. It allows real-time visualisation of cell behaviour, interactions, and multicellular structures. Researchers can explore various tissue types, from trabecular meshwork to liver, breast tissue, stomach, ovarian tissue, and even the central nervous system (CNS). By maintaining physiological conditions, including temperature, pH, and oxygen levels, IVTech’s LiveBox1 bioreactor provides an accurate environment for studying drug efficacy, toxicity, and disease mechanisms.
LiveBox2 is a transparent bioreactor that mimics pathophysiological barriers in dynamic conditions. It supports both air-liquid (ALI) and liquid-liquid (LLI) interfaces. Researchers can directly interact with tissues during experiments using the ALI module. LiveBox2 is suitable for maintaining spheroids in floating conditions and enables forced perfusion conditions.
MultiDyn is a compact multi-dynamic-chambers rack based on standard 24-well plate dimensions. Each chamber can be autonomously perfused, allowing for 4 parallel and dynamic models. It enhances compatibility with LiveFlow, providing flexibility for various experimental setups.
IVTech’s bioreactors have been successfully used with 2D or 3D cell cultures, including bioprinted tissues, floating or attached spheroids/organoids, and animal or human biopsies. Researchers can test drug effects on specific targets and related tissues, increasing the predictivity of in-vitro models. Additionally, biological circulating material, such as lymphocytes, can be injected into the IVTech system, further enhancing its human-relevant capabilities.
Peristaltic Pump for Dynamic In-vitro Modelling
IVTech’s LiveFlow peristaltic pump is an advanced tool for dynamic in-vitro modeling compatible with incubator environments. This compact and lightweight pump incorporates two independent motors, allowing researchers to simultaneously implement dual flows with varying flow rates or patterns for diverse experiments.
LiveFlow stands out with its ability to replicate the interstitial flow of real tissue. By mimicking interstitial flow, LiveFlow facilitates the study of fluid dynamics within tissue-like environments, providing valuable insights into drug transport, cellular responses, and tissue interactions.
Designed as an all-in-one platform, LiveFlow includes a drawer housing the fluidic circuit. This allows for easy management of modules when transferring systems from the cabinet to the incubator, enhancing usability and efficiency in research settings. The compact size ensures efficient use of lab space within the incubator, allowing researchers to seamlessly integrate LiveFlow into their experimental setups.
Accurate Disease Modeling
LivePa is a pressure modulator critical for researchers aiming to create in-vitro models that closely mimic the onset of certain pathologies.
By enabling researchers to increase hydrodynamic pressure, LivePa simulates phenomena correlated with pressure changes. These include disease-related conditions, such as hypertension, glaucoma and tumours, as well as physiological adaptations due to environmental stimuli, such as increased pressure during food ingestion, supporting accurate disease modeling. The flexibility to set up different rates of pressure increase allows tailoring experiments to specific conditions.
LivePa seamlessly integrates with the incubator environment, making it a practical addition to experimental setups. Placed into the LiveFlow drawer, it works harmoniously with other components. LivePa is compatible with both LiveBox 1 and LiveBox 2, ensuring versatility across different research contexts.
